In this talk I will present our work on the preparation of macrocyclic and acyclic CB[n], studies of their fundamental molecular recognition properties, and their use in biologically relevant applications.  For example, I will discuss the: 1) Ultrahigh affinity of CB[n]•guest complexes, 2) acyclic CB[n] as solubilizing excipients for insoluble drugs and as an in vivo reversal agent for neuromuscular block, 3) monofunctionalized CB[n] for targeted drug delivery, and 4) metal-organic polyhedral functionalized with CB[n] groups for drug delivery applications.
